EDIT: to elaborate my previous response, what I meant was that saying 5th was used was because that was closer to what people see in the real world in terms of engine load, is dependant on where that person does most of their acceleration on that car? The rolling resistance in the dyno should present the load that the car would see under normal driving conditions, so if that person doesn't do most of their hard acceleration in 5th, then tuning in 5th might not most closely reflect what that persons car sees in the real world, i.e. while that might be against maximum load, if the car isn't generally run against that maximum load, then that isn't the best 'nominal' load against which to tune it?
That's my thought process 'out loud' based on understanding engineering fundamentals though, not being presented as 'fact' or on experience in actually carrying out engine tuning...
